Ah man, mutants? Well I don't like the sound of that too much. When I watched the first trailer it bought The Stand to mind, in which events were kind of similar - 99% of the population being killed by a virus. But rather than dying they're being 'mutated'? Not sure how I feel about that. Also 20 years, man thats a long time. I think it was in a docu I saw once, which surmised that without many people around to stop weeds and grass and the like from overgrowing, urban areas would be reclaimed but nature pretty darn quick post-organised civilsation. So I didn't think it'd be that long.
I mean, I know that they have carte blanche to do what they like, it's their post-apocolyptic world and all...but basing it more in reality, and struggling to survive against other aggresive humans would be much better. Mutants are a bit too fantastical for me, in this setting anyway. Also, if no one dies from the virus, and they're all 'mutated'...will they be all wondering around together, like (dared I say it) zombies?
Then again, the mythological and kind of paranormal stuff in the Uncharted series worked pretty well. Apart from those random spirit things at the end of 3, but I'd rather not think about them.

They're basing it off some fungal infection (cordyceps i think) like 28 Days Later meets I Am Legend and mad broccoli infection. They said the focal point is not the 'zombies' but more on the story and relationship development between the dude and the girl and their fight for survival, so I think you'd be happy there!
